CRISPR(clustered regularly interspaced short palindromic repeats) is an integral part of modern genome editing processes due to its versatility. However the usage of CAS(CRISPR associated) protein blockers is required in some applications. Designing structures such as macromolecules is not a simple process, it requires a lot of resources, alongside people competent enough to work with it. Fortunately, there are some examples of natural protein blockers that we can use in our applications, such as AcrIIA1. AcrIIA1 function is connection to biochemical bonding sites, preventing DNA from being destroyed by the CRISPR immune system. By analysing AcrIIA1 we may learn more about natural protein blockers and possible simplifications while developing artificial blockers for other proteins.
