To date the critical importance for quantitative investigation of biotherapeutics and their targets in tissues has been fully appreciated. However, tissue analysis remains challenging owing to the low tissue levels, the high interference and the lack of appropriate sample treatment methods achieving sufficient quantitative accuracy and robustness. This lecture will elaborate on the technical difficulties associated with tissue analysis and then the analytical techniques we developed to improve sensitivity, selectivity, accuracy, throughput and robustness of LC-MS-based tissue quantification. A number of applications will be given as examples, such as i) sensitive and comprehensive tissue PK analysis, ii)quantification of antibody-drug-conjugate, free- and conjugated toxin in preclinical models, iii) study of tumor penetration and immune cell recruitment by T-cell redirecting bi-specific antibodies and iv) accurate quantification of the intra-tissue distribution of mAb and target using 3D-printed micro-sampling devices.
