In this presentation we will describe a novel modeling approach incorporating computational fluid dynamics (CFD) and physiologically-based pharmacokinetic (PBPK) models to simulate the deposition, absorption, and bioavailability of nasal corticosteroids. CFD models of the human nasal passages were used to estimate the droplet deposition locations of nasal sprays. PBPK models were developed to simulate the subsequent nasal tissue disposition after droplet deposition, absorption through the nasal mucosa, and availability to the systemic circulation. The PBPK models incorporated local nasal deposition patterns, mucociliary clearance, dissolution, tissue diffusion, receptor binding, plasma protein binding, metabolism, and GI clearance. Results will be presented for three commonly used nasal steroid sprays.
